Expressing recombinant human lactoferrin with antibacterial activity in Nicotiana benthamiana
نویسندگان
چکیده
Lactoferrin is a non-hematic iron-binding 80-kDa protein that exhibits antimicrobial activity. Higher plants function as “green bioreactors” for large-scale recombinant production. In this study, we transiently expressed human lactoferrin (rhLF) in Nicotiana benthamiana at yield of approximately 40 µg g−1 fresh mass (gFM) using the Tsukuba system. Additionally, expression level rhLF increased when it was fused with KDEL, an endoplasmic reticulum retention motif. Purified plant-derived possesses antibacterial activity inhibits growth Escherichia coli. These results indicated containing can be produced N.

Antibacterial activity in bovine lactoferrin-derived peptides.
Several peptides sharing high sequence homology with lactoferricin B (Lf-cin B) were generated from bovine lactoferrin (Lf) with recombinant chymosin. Two peptides were copurified, one identical to Lf-cin B and another differing from Lf-cin B by the inclusion of a C-terminal alanine (lactoferricin). Two other peptides were copurified from chymosin-hydrolyzed Lf, one differing from Lf-cin B by t...

Lactoferrin (Lf) is an iron-binding glycoprotein which exhibits antibacterial activity against a broad range of Gram-negative and Gram-positive bacteria. The mechanisms of this property are complex and involve iron sequestration as well as direct interaction with bacteria. Lactoferricin (LFcin), a Lf-derived peptide released by pepsin digestion of lactoferrin, is a more potent antibacterial com...
متن کاملAntibacterial activity of lactoferrin and a pepsin-derived lactoferrin peptide fragment.
Although the antimicrobial activity of lactoferrin has been well described, its mechanism of action has been poorly characterized. Recent work has indicated that in addition to binding iron, human lactoferrin damages the outer membrane of gram-negative bacteria. In this study, we determined whether bovine lactoferrin and a pepsin-derived bovine lactoferrin peptide (lactoferricin) fragment have ...

Lactoferrin (Lf), a member of the transferrin family of iron-binding proteins, is now known to have a number of properties, including antibacterial activity towards a broad spectrum of Gram-negative and Gram-positive bacteria. The mechanism of the antibacterial activity of Lf is complex and involves beside iron-chelation, direct action on bacteria and/or the activation of the immune system. Lac...
متن کاملAllergenicity Assessment of a Genetically Modified Protein-Recombinant Human Lactoferrin
Introduction Human lactoferrin (hLF) is known to be a multifunctional protein of the transferrin family. It is a glycoprotein of approximately 80KDa and is present in various human secretory fluids including milk, saliva, tears and nasal secretions as well as being abundant in neutrophils [1-4]. The hLf protein includes 703 amino acids as a single polypeptide chain that folds into two similar g...
